PI3-Kinase Class 3 (VPS34) is the catalytic subunit of the PI3K complex. It is involved in the transport of lysosomal enzyme precursors to lysosomes and the induction of autophagy during nutrient deprivation (1, 2). Thus, PI3-Kinase Class 3 (VPS34) regulates intracellular vesicular transport at multiple steps through the production of phosphatidylinositol-3-phosphate (PI(3)P). Certain downstream effectors are involved in each of the processes that PI3-Kinase Class 3 (VPS34) regulates. It exerts its function through the recruitment of proteins that contain PI(3)P-binding motifs such as FYVE or PX domains (3). PI3-Kinase Class 3 (VPS34) is also required for the activation of the mTOR (mammalian target of rapamycin)/S6K1 (S6 kinase 1) pathway, which regulates protein synthesis in response to nutrient availability (2).
