Mammalian cells are the preferred expression platform for proteins requiring fully mammalian post-translational modifications. As such, they are commonly used for the production of antibodies and therapeutic proteins. They are also preferred when the downstream application for a recombinant protein is in a functional mammalian cell-based assay. At NeoBioLab we produce proteins in both transient and stable formats using HEK293 or CHO cells. We utilize proprietary expression vectors to drive high levels of expression and regularly return >10mg/L of recombinant protein. Our superior capacity allows us to conduct tens of transient expression projects simultaneously and our serum-free suspension culture format allows for large-scale production yielding grams of the desired protein.
We can take a customer provided cDNA or optimize and synthesize any desired coding sequence and subclone it into a mammalian expression vector. Once cloned, all constructs are restriction digest and sequence verified.
Generally, we conduct small scale expression and purification trials to identify the best clones and/or expression conditions along with optimal protein isolation and purification procedures. We use these conditions to produce approximately 100 micrograms of protein.
For larger protein requests, we scale up expression using optimized conditions. We utilize 20L Wave Bag systems to produce gram quantities of the desired protein.
